i left them charged overnight and there is a mild buzzing. Is it bad to leave it like that? Will it do damage to the membrane?

Even if it were to damage the membrane, you destroy the membrane during a rebuild anyway. so if you can't get the dirt out of it with vacuuming and compressed air, then the panel is toast and who cares of the membrane gets damaged.

If you have a high voltage leak, it may pull down the power supply and make the whole speaker slightly more quiet.
