Oil based Kilz use multiple lite coats and full dry out between coats... including screw holes, should work well. Best if applied with a gun.

Anything non-porous will seal MDF. Dip it in old sump oil, if you want.
I use latex-based acrylic house paint, myself. Ridiculously cheap.

Dave Pelegrine has shown trench cutting the seams and fill with auto body filler works the best long term. Do a search here on diy for more info.

The intent is to make it air-impermeable. Yes, sump oil will work. Aesthetically, not the best, but it is air-tight.

A LAB12 in a ~40L sealed box will need some EQ to get low. OP, do you have any particular amplifiers in mind for the subwoofers?
A cheaper option is a JBL GTO1214, or the Infinity equivalent. The T/S parameters are different to the LAB12 etc, but when you're using EQ anyway, its more the distortion performance of the driver that's of interest.

The point is to make it impervious to moisture otherwise it will swell.
