I am newbe in this subject. I would like to remove a 100k pot in my 300 B amp. Do I need to replace it by 100 k resistor?

I am not sure. All I know that this pot is between first and second stage.

You do not absolutely need to add another resistor, the 1 Meg resistor R21 is already there. The first stage gain will go up slightly.
Personally I would not literally remove the pot, but just wire the top of the pot to the wiper, ie connect C9 to R15 and R21. That way you will have less work to do if you change your mind.
