I'm building a pair of wmtw towers. Drivers are Beyma 15lex1600nd, 12p80nd and JBL 2451 in a JBL 90x50 waveguide. Om my testboxes i have the slots for the 15 inchers located horizontally below and above the drivers in each end of the baffle. On my final boxes however, I'm thinking of placing them vertical on each side of the 12" and waveguide like on the Genelec 1032.
My concern is how it will affect the sound? The bass wont get additional support from floor but what else?

Depends on how wide the vent's BW, but usually the WLs are so long that it doesn't matter. To be sure though, find the port's highest audible frequencies' 1/4 WL (~34400 cm/4/f) to see if it meets this criteria for summing as one, i.e. acoustically doesn't matter, though how it interacts with the room's acoustics are more likely to be audible as it might move it from a peak to a null at the extreme 🙁, requiring finding a different speaker vertical height and/or tilt, which in turn might no longer be optimal for the main listening position.
Now you know why I'm no fan of tall multi-ways unless sealed and even then better overall to do an infinite line array IME.
