The accuracy of the Spice simulation is directly related to the accuracy of the models.
Some models are better than others. I've had problems with screen current not modeling properly at low currents. Other than that, most models I've found on the web are pretty good.
Spice is a nice tool for playing 'what if', but it will only take you so far before you have to build the circuit. Simulations can point out gross errors, but don't really tell you anything about how it sounds if there is nothing really off.
I made a model of the 6P41S using a java program from :

I thought it was totally screwed up as it showed grid bias at -32V. When I built the amp, I was surprised to find the model was very accurate.
My 6DX8 model wasn't as good.

Apart from audio waveshape, I mainly use TrueRTA for instant frequency response, and spectrum analysis.
You can make a pretty simple above-audio oscillation checker by hpf, rectifying and filtering for a dc level.
TruRTA can't do X-Y plot very important drawback IMHO.
