Is it possible to reverse driver/port location on a MLTL enclosure and get exactly the same performance, without retuning the port and change stuffing density ?

Pretty much, but not exactly the same. There is a bit of difference in the frequency reponse between the first and second resonances. Test it for yourself in MJK's mathcad worksheets. I built my speakers as a mass-loaded "reverse" voight and they modeled very nicely (and realised quite nicely as well).
