It could restrict the bass performance, anything else?
Without getting into the arcane and oft argued area of frequency distribution of edge diffractions, yes compromised bass response would likely be the first factor that is to judge the efficacy of a such a design.
As for the rest, time for refilling of bladders, and let the water-sports begin.
We are still left with boundary gain and missing fundamental though. Regards the room, I have a nice big mode at 40Hz which is handy 😉 There is still room gain below it's fundamentalOf courset depends, the room dominates below ~300 Hz, but the F3 does not play into that.

My room is large enuff, and complex enuff, with a high vaulted ceiling and i have few issues with the room. There are some modes, but the only noticable one is in the far corner of the kitchen, and only with really low that usually is not in th music.

Here’s an internal pic with the foam out. I’m going to use layers of felt instead of the foam.

Just put the drivers in! Sounds pretty good. Maybe a bit bass light.

What else should I do to optimize?

As I listen more I am really liking it... why does it sound so good without any adjustments?

Wait 100 hrs.
Did you suss out a closer estimate of the box volume. What is the existing vent?

Gee, maybe there’s something to this single driver thing after all?

I am really happy with the sound so far. I’m not really sure about the exact internal dimensions beyond what I’ve already posted. Now that the drivers are in I would rather leave them alone. The bass is light but it’s not a problem for me. I just want to be sure that the boxes I’m using aren’t distorting the sound in an undesirable way. I just don’t really hear that. Thanks to everyone who contributed to this thread.
Bass light could well mean the box is tuned too low for the FR. No easy way to sort that but shortening the vent.

It would have been probably easier buying a kit for a dedicated driver like Fostex, the result wouldn't have been a headache.

Makes sense. Looks like the Dayton 6.5” woofer has a “free air resonance” of 34Hz while the 10p is 42Hz. Seems logical since it’s smaller. I would imagine shortening the tube could help as you suggest.
