If you were going to use more than one driver, do you multiply that 1-1.25 liters by the number of drivers ?
Yes.
or would it be better to build the cabinet with separate divisions for each driver?
Separate division for each driver is not necessary. But, if you are using many drivers (e.g. line array), box may be too long (tall) and produce internal standing wave (resonance), so in that case it is better to divide the box in several divisions, with up to 6 drivers in each division.
 
Thanks guys. Mind if I take this farther?
I'm going to try throwing something together with scrap ply - a single mini tower with three drivers facing forward and six in a slot below. The slot loaded six will be three facing three. Do I want to have each of the three sets of three drivers (the forward facing three above and the pair of threes in the slot) in its own volume?

Also, how does larger volume affect FR? Should I adjust the volumes to optimize for HF and LF (slot) sections?
