You don't usually use the term "gain" with power tubes - it make sense with voltage amplifiers, where it's the ratio of voltage out / voltage in.
The tube data sheet will give the grid signal required for a certain power out - find 'em here
6V6 is usually operated at 300V or below for class A - at 40 mA it is right at its dissipation limit of 12W. 6L6 can be operated up to 360V at about 55 mA (19W limit) - the 6L6GC can go higher, but it's better suited for AB operation.
