What is the best PVC port to solid wood glue?
Maybe PVC glue to melt the PVC a little and then some other glue to go with it?
Or is there a glue that is sticks well to PVC and wood?
I figure just about anything glues wood since it's so porous.

I use a thick bead of hot melt on the inside of enclosure - silicone would work as well, but can be messier, and takes longer to cure

Definitely don't do that.
Epoxy would be great for this and if your hole wasn't perfect it will also gap fill.
I used Liquid Nails, available at Home Depot and Lowe's, to glue the PVC ports depicted in the post linked below.
