I'm building a P3A amp that will be paired with the ESP tone control preamp. The pre already has an output cap so I'm wondering if there is any reason to keep the input cap on the P3A, and if not, could I also eliminate R1, R2, & C2?

You might remove it but R1 and C2 is another filter (low pass), don't remove it.
Also since your tone control has 1uf cap you might suffer some bass loss (system dependent) , if possible use a larger value like the suggested one for P3A, 4.7uf for C205/C105.

The input cap, 4.7uF is large enough. If it is electrolytic type capacitor, you could swap it to film or ceramic type capacitor.
For the preamp, I would remove C105/C205, R115/R215.
