I want to attach my front baffle to an MDF box but make it so it's a good fixing that can be undone and done up again.
Can anyone recommend a method for this?

MDF is very easy to strip with repeated removal / re-installation of screws (often more than twice is too much) If you want to be able to repeatedly install/ remove driver baffles, I'd suggest threaded inserts - NOT T-Nuts - as well as a perimeter gasket of closed cell weatherstrip tape.
