I Think I misunderstood the description at the Whispermat homesite, mentioning a "barrier septum"..

I think that part of the confusion is because they are describing how their product is used in say a factory where the noise is coming from the inside of a cabinet or duct and the material is used to quiet it for people who are in a room.

Whispermat doesn't contain structural foam either,
IMO the basic sheet is the same as using vinyl floor tiles.

However IMO a suspended vinyl layer between sheets of
foam would not work well on the scale of speaker cabinets.

I agree that the vinyl layer in the Whispermat is aimed at doing the kind of thing that vinyl tiles in speakers do. However, the Whispermat vinyl is a high density vinyl and is noticably heavier than most of the vinyl tiles I have picked up. This makes sense since a vinyl tile is often meant to be somewhat "cushy" when walked on.

So I think it is a matter of degree. Vinyl tile will have an effect above straight MDF. Whispermat likely has a still higher higher mechanical damping effect.

I think it is a matter of degree. In any given application, straight MDF may be enought, MDF+vinyl tile may be needed, or MDF + Whispermat/Blackhole may be prefered as a no compromise solution.
