Some additional details would be really helpful, things like what it is connected to, some history such as whether it is new to you or previously worked and has now quit, whether or not you have checked the source (sources), speaker wiring and presence of AC power. (apparently it powers up) What has changed since the last time you used it?

Measure DC voltages between ground and each pin of the 300B. Repeat with the driver tubes if you see more than 300V on pin 2 and more than 50V at pins 1 and 4 of each 300B socket. (If you see 300+V at pin 2 but less than 50V at pins 1 and 4, measure pin 2 and look for negative voltage)
"Good" and Radio Shack don't compute.. LOL So you have an inexpensive meter, and probably are not getting a valid resistance reading of the secondary winding.
What is resistance measured when you short the meter leads together? I'm betting it is a long way from zero.. (hopefully)
