isolating mounts for transformers

This isolation works as a mechanical isolator. It prevents the whole chassis from humming together with your transformers. Good thing to do and relatively simple.

I have tried earthing of transformers and chokes but I really can't hear any difference.

What is the purpose of this? Is it safe not to have the case of the power transformer connected to ground? What about any shielding provided by the case?

No problem.
I'd recommend rubber washers or similar to control xformer / choke induced vibes from entering the main chassis.

When not tightened too hard simple rubber grommets can be quite effective.
