So the first subwoofer I'm trying to build ever is going to use a Dayton Audio SD315-88 12" subwoofer and a Foster WF-100K plate amp. I dug up some old documents on the amp and it states, "When VR603 is turned fully counter clockwise, the spectral tilt is almost completely removed. If you want the spectral tilt 100% gone, you will need to jumper a 10k resistor across pin 1 and 3 of VR603."
I suppose I have two questions.
1) How would you describe spectral tilt to a total newb?
2) For building a sub that is intended for music, would you suggest following the guide to completely remove spectral tilt? Will it even make a difference?

Depends on room acoustics, where the sub is located and its roll-off slope.
Ideally we want a tonally balanced response ['flat' to our ears/brain] from the lowest lows to highest highs at least at the listening position [Lp].
