Testing stuffing

I have read people making references to testing out different amounts and kinds of accoustic stuffing in their speakers.

How exactly do you go about this? Do people leave one side of the speaker unglued until they have finalized the damping? Doing it through the hole for the driver seems unlikely in a TL configuration.

I make the top of the boxes removable. That's the easiest for me.
Rope caulk makes a good seal.
