Checked several times, seems to me they are correct: green to RXD and white to TXD.

Are you using the ISO RXD and ISO TXD pins on J3 header for this? These are for TTL level, the manual really needs to explain this better.

Someone also mentioning a problem using that blue interface with dam1021 here, isolated input expects 3.3V levels, could it be outputting 5V levels?

I have used J10 RXD-TXD.

J10 is for rs232 so that would be the problem.

Now I get the correct response but there is no way to start the uManager.

Do you have the rev7 board with TTL-RS232?
I once had similar experiences with a mainboard with TTL-RS232 ports. I had to invert the levels to get the correct output (the logic levels of real RS232 are inverted).

Then it is clear you have the wrong USB adapter. You need one with a real RS232, for example this one:

The one you have pointed out uses the same Prolific PL-2303 Chipset, so I don't think it will solve the issue.

Chipset doesn't necessarily matter, voltage and polarity matter. My understanding of serial comms is that the same chipsets can run both 3.3-5V TTL and 'proper' ±13V RS232 depending on the surrounding circuitry.
