The answer to that could fill a book. Or at least a chapter or two of a book. A few days spend studying the Radiotron Designer's Handbook will help a lot- this is a complex issue with a lot of variables.
Or... you can just go with the recommendations of the tube manufacturer. Most data sheets for power tubes give suggestions for load, bias, power, and distortion.
