Irrelevant questions: the amp is inverting. You need non-inverting. That means, you need to go with 2 stages and feedback around them defining voltage gain.

all your questions are redundant

regulator is not having any connection with "if I don't use DN2540"

in this case , Iq of stage is set with DN2540 at 5mA , so Salas reg must nbe set at at least 10mA , twice that even better

anyway - if you're going to use proper shunt reg - taking in account that reg itself demands some space , then use proper tube ....... with lower Ri , greater xconductance

I don't like sissy tubes

6Н23П (ECC88=6DJ8) is weakest tube I'll use in line stage

so - 3 stages - two gain stages , cascaded , with cathode follower

look for any of Allen Wright's SE line stage constructions

edit : didn't saw x60 ......

in that case AW constructions aren't helpful ......
