I think it's evident that the mentions of leaving B+ and DRV alone for off-board Power Drive, etc means that the 'normal' build jumpers these, but I want to make sure.

DRV_B+ gets a jumper, yes. I believe that you leave off this jumper if you want to isolate the FET drains from the B+ rail so that you can use them to drive a different pair of tubes running on an external B+ rail.

Actually I would be very interested in using the board as a driver board. I was thinking of trying to use the 45 to drive the 300B. Would it work ?

It can drive 300Bs directly in the on-board sockets....
