You are talking about a large room and a smallish subwoofer, if you cannot find adjacent room for an in-wall / infinite baffle installation you will have to make a few compromises.
For starters: it would be good to know what your required listening levels are; e.g.: for music? home theather? background?. The Radio Shack sound level meter or any inexpensive sound level meter would be very helpful:

Your desired final size ends up to be roughly a 15-3/4"[40cm] cube, and I assume that means the finished size (including grill and recessed amplifier mounting). That leaves you with a volume that is generally too small for a 12" subwoofer. One possible solution would be to build something around the CSS SDX10 10" sub. You could start with a sealed construction to fit your size requirements, and if you want additional output at the low end you could try a size modified version of CSS's Quartet10 kit:

This adds two passive radiator to the active driver, and will provide much higher output at the low end. For modeling a passive radiator loudspeakers I recommend Jeff Bagby's free software (Excel app.):
