How to Calculate volume of enclosure for a mid-range driver?

I know it is able to calculate the volume of the bass driver from its T/S parameters. Closed or Vented.

How about the mid-range driver if I cross it at 250Hz(High-Pass) in a closed box?

Or it is not so important because it just deal with the frequency above 250Hz?

As long as there's no bump in the frequency range you'll use the speaker in (or just outside), then you should be fine.

I usually design it to always have a rising excursion at lower frequencies, so no hump in the excursion graph. Preferably smooth in appearance.
I also like total Q to be lower than 0.5

you can model this in Unibox, it is important because it affects the fall-off rate & this interacts with your crossover slope. However it's not critical unless the box is too small, when you start to get an LF hump.
For me, the internal damping & shape of the rear wall are just as important, to avoid internal reflections
