There are two triangular chambers in the B'horn Mk2 design. I see mention of filling the bottom chamber with sand but nothing about the upper chanber. Should both be filled? I realize the sand in the bottom will also make it more stable. However, is it necessary for the upper and has anyone ever used an expandable foam like Good Stuff expanding spray foam?

The upper cabinet is usually stuffed with poly-fil or wool. It may not be wise to leave it empty as it would become a resonant chamber.

Have a look at the zHorn site, where he recommends filling voids with some kind of architectural foam.
