If using two woofers in an enclosure calling for 28 liter each volume, does it make a difference sonically if you construct the enclosure as one unit multiplying the volume by two...or should you separate each 28 liter area internally?

Most studies have indicated that it's not an issue whether you build them separately or together, as far as performance is concerned. I'd build them separate for a couple reasons. The separator is going to act as a shelf brace, there will be less concern with box modes as the (likely vertical) 1 dimension will be halved, making a higher frequency resonance that's more easily damped, and also, in a failure situation, you'll have more airspring protection on the remaining driver if one blows.
