The removable back is a design feature I put into the pencil cabinets to help people tailor the damping to their particular requirements.

Yes, Alpair 6P is 3-inch paper cone full range driver.

Once stuffing has been tailored to the end-user preference, is there anything to gain by sealing , as in making air-tight, the cabs?

Around the removeable back you mean? Yes, it should have a good seal. A strip of rubber sealing tape or similar around the stringers should do the job nicely. Difference won't be vast, assuming the box is made with a tight tolerance, but it's worth doing.
